低溫電顯核心實驗室

  • 歡迎來到中央研究院低溫電顯核心實驗室!



    低溫電顯技術最主要的優勢為可解析生物巨分子之結構在最接近生理之狀況。

    為了要製作適合低溫電顯觀察的樣品,生物樣品須以小於微秒的時間瞬間冷卻至液態氮溫度,並將生物分子冰凍在非常薄之無序冰中。

    因低溫樣品製備技術能夠直接攝得固定在無序冰中樣品的暫態,此技術亦具有解析巨分子動態的能力。 再者,低溫電顯斷層攝影術(Cryo Tomography)可較為快速獲得生物巨分子之結構。


    本設施主要服務項目為低溫樣品之取圖與三維重構等。 本設施可依使用者需求,針對大於200kD 的蛋白巨分子、病毒等提供服務與技術諮詢及支援。

    服務流程簡述如下:

    1. 低溫樣品製備:預約討論時間 → 討論或指導樣品製備流程 → 樣品製備
    2. 上機服務:由專人委託協助上機。欲自行上機操作者,須經過儀器管理人認可方得自行上機。
    3. 資料擷取與運算:包含影像取得與分析運算、3D結構重建等。
