有絲分裂期細胞崩猝是一種細胞於有絲分裂期死亡的形式,通常導因於DNA損傷的誘發或有絲分裂的異常,目前被認為是許多抗癌藥物達到療效的主要機制。另一方面,遺傳不穩定性是腫瘤細胞的特徵之一,其誘發也是導致細胞癌化的重要原因。核酸、染色體、及控制染色體分離的系統是許多致癌劑所攻擊的標的;而核酸損傷或染色體異常的誘發或干擾染色體分離常會導致遺傳不穩定性的發生。因此,在有絲分裂異常的細胞中,若細胞崩猝無法被適當的誘發,不正常的分裂將有可能導致子細胞發生遺傳不穩定性。

利用會造成DNA或紡錘體損傷的化合物,我們在細胞的層次探討這些損傷如何誘發分裂期異常,以及分裂期異常如何誘發細胞崩猝。這些結果將有助於相關疾病的預防或治療。目前的研究主題有:(1)了解有絲分裂期細胞崩猝的分子機制;(2)探討磷脂醯肌醇(phosphatidylinositol)在有絲分裂進行時對微管動態(microtubule dynamic)和細胞抗藥性的影響(Figure 1);(3)篩檢促進有絲分裂期細胞崩猝的新穎化合物(Figure 2)。
